How long do agar plates last? Plates will keep refrigerated for weeks. How do you heat up Agar? Loosen the bottle cap, but do not remove the cap while heating. Warm the agar bottle in a hot water bath or in the microwave on medium for about one minute or until it becomes liquid. What would happen to plates poured with agar that is too hot? If plates are poured with agar that is too hot, then the agar would kill the bacteria.

Agar is the support medium for bacterial and fungal culture, as nutrients, antibiotics, salts and various growth enhancers. Too hot agar would kill the bacteria as the bacteria would get denatured.

How do you clean an agar bottle? You can clean it by filling it with water and microwaving it until it boils. Boiling water will dissolve the agar. It is done right after dishes are poured.

Thanks folks! This will be handy to know for the future. Now my only complaint is that our stores only have packs of 20 and 30 sterile plates which is great for doing a class set but a pain in the butt when I only need a handful, it seems such a waste the plates arent thrown out of course they are kept for other uses but I have about a hundred non-sterile plates that I'm struggling to find a use for haha. Keep the plates in the bag an seal as soon as possible.

Very unlikely they will become contaminated. Hello BioBee, I keep the unopened plates in the bag and reseal it - they are still sterile. It'll be fine for school use to re-melt it. Interestingly though, we never did this when I was working as a research microbiologist. The sugars in the agar caramelise when it's autoclaved hence why some agars go a bit darker on autoclaving and these combined with heat can result in inhibitory substances being produced.

As I said, this is not an issue with school level work, but we were looking at sublethally injured organisms, and they often wouldn't grow if the agar was heated more than once! Yeah, you might've misunderstand the instruction somehow. The usual packet of agar powder here, which is 10g packet, is supposed to be dissolved in a litre of water and a cup of sugar to get a bowl of agar dessert.
